Vimal Yoganathan had given the Tykes a seventh-minute lead but Lisbie pounced in the 54th and 68th minutes to secure a hard-earned home victory.
The lowly Tykes, who entered the game winless in nine in all competitions, snatched an early lead.
Skipper Cameron McGeehan forced a fine stop from home goalkeeper Joel Pereira but, in the ensuing goalmouth scramble, Yoganathan scuffed in a shot from close range.
Pereira also had to save smartly from Tobi Adeyemo’s fierce drive before Reading finally awoke from their slumber, with Jacob Brown testing Barnsley keeper Cieran Slicker with a close-ranger header.
Reading continued their momentum after the break, levelling through Lisbie after a Randell Williams cross from the left had fallen nicely for him at the far post.
Lisbie completed the turnaround when he smoothly guided home past Slicker from a Josh Stokes pass.
Barnsley threatened a late equaliser when Reyes Cleary broke clear but he drove wildly across the face of the goal while Lisbie was denied his hat-trick by a superb stop from Slicker in stoppage time.
