The pair are amongst the 37-strong squad named by fellow Borderer head coach Gregor Townsend.
Scotland host New Zealand (November 7), Australia (November 15) and Japan (November 21) before taking on an as yet unknown opponent at Twickenham in London at the end of the month.
Gala's Townsend had delayed naming his squad for a week in the hopes of allowing several key players the chance to prove their health.
Graham, Scotland’s leading try scorer, was initially ruled out for the remainder of 2026 following a bicep tear in pre-season. But the 29-year-old featured for Edinburgh in their 27-17 European Challenge Cup loss to Toulon last weekend before being dropped as a precaution for Friday's United Rugby Championship win over Lions.
Prop Sutherland, 34, received the nod after starting Glasgow’s 24-20 win over Benetton at the weekend on his return from a calf injury.
Scotland head into the November fixtures sitting second in the Northern Hemisphere table of the inaugural tournament, having won two of three fixtures earlier this year in defeating Argentina and Fiji either side of an entertaining 42-28 loss to South Africa.
